This particular disease is the model for emphasizing the importance of basic research in obtaining practical solutions to clinical problems. Diphtheria was first identified as a specific clinical entity in 1826 after the publication of Pierre Bretoneau's classic monograph, but its bacterial etiology was not established until 1888. Klebs had earlier described the characteristic bacilli in the pseudomembranes of diphtheria patients and Loftier had isolated the organism in pure cultures, but the full understanding of the pathogenesis of the infection was obtained only with the discovery by Roux and Yersin of a soluble exotoxin in culture filtrates. From there, continued study until the discovery of a toxin and a toxoid, substances that have been used in active and passive immunization against the disease.La difteria es el prototipo de una enfermedad toxigénica, o sea una infección aguda causada por cepas de corynebacterium. Esta enfermedad en particular es el modelo para notar la manera de enfatizar la importancia de la investigación básica para obtener soluciones prácticas en problemas clínicos. La difteria se determine como entidad clínica especifica, por primera vez en 1826 luego de la publicación de la clásica monografía de Pierre Bretoneau, pero su etiología bacteriana no se estableció hasta 1888. Klebs ya había descrito más tempranamente los bacilos característicos en las pseudomembranas de pacientes diftéricos y Loftier había aislado el microorganismo en cultivos puros, pero la total comprensión de la Patogenia de la infección se obtuvo solo con el descubrimiento efectuado por Roux y Yersin de una exotoxina soluble en los filtrados de los cultivos.
